Biology 376 Links for unc-86 - POU domain proteins lab

C. elegans unc-86 + POU-domain proteins + terminal selector lab GROUP TASKS

Most can be done in any order - so while one group is using the microscope, others can do some of the computer-based tasks
  1. Prepare a multiple sequence alignment of C. elegans unc-86 (top) with other nematode unc-86 orthologs (or at least POU domain proteins), plus human Brn3b and POU4 class proteins.
  2. Photograph a z-series of at least one high quality stained adult or larva of your species of interest (either Oscheius tipulae [strain CEW1] or Pangrolaimus sp. [strain PS1159]. (Dr. Loer will assist in getting you started, and selecting a high quality specimen from the slide.)
  3. Transfer your images to a separate computer. Download FIJI and use it to convert your images into a stack [Images: Stack: Images to Stack]. Play with brightness & contrast to optimize image.
  4. Determine the pattern of expression (make a schematic image) for your species of one larva and one adult. (There are some already prepared stacks online.) Compare to the C. elegans pattern of expression (see schematics in the resources folder), and speculate on how they are different - can you identify any likely 'homologous' cells in the other species?
  5. For next Wednesday (Dec 11), prepare a brief presentation with your group with an introduction to terminal selectors, your nematode of interest, unc-86, and then show your data and results - alignment(s) and pattern of expression in an adult and larva. (Inculding compare/contrast with C. elegans)
